Research Progress of Targeted Interferon Regulatory Factor 5

  • 摘要: 干扰素调节因子5(interferon regulatory factor 5,IRF5)作为一类关键转录因子,在调节人体免疫系统功能方面具有重要作用。通过调控细胞因子与趋化因子的表达,IRF5能影响多种免疫细胞的炎性反应过程。多项研究结果显示,IRF5的功能失调与系统性红斑狼疮(systemic lupus erythematosus,SLE)、肌萎缩侧索硬化症(amyotrophic lateral sclerosis,ALS)等多种炎症性疾病的发生发展密切相关,同时在肿瘤的恶性转化与免疫调节过程中也起重要作用,并已成为药物研发领域的一个备受瞩目的治疗靶点。然而目前对于精准干预IRF5途径的药物研发仍充满挑战。综述深入探讨IRF5在调节免疫细胞功能中的重要作用,并对现有靶向IRF5的干预方法进行系统回顾,以期为未来开发用于治疗免疫性疾病的IRF5抑制剂提供参考。

     

    Abstract: Interferon regulatory factor 5 (IRF5), as a class of key transcription factors, plays an important role in regulating the function of the human immune system. By regulating the expression of cytokines and chemokines, IRF5 can affect the inflammatory response of a variety of immune cells. Many studies have shown that the dysfunction of IRF5 is closely related to the occurrence and development of a variety of inflammatory diseases such as systemic lupus erythematosus (SLE) and amyotrophic lateral sclerosis (ALS), and that IRF5 also plays an important role in the malignant transformation of tumors and immune regulation. It has become a high-profile therapeutic target in the field of drug research and development. However, it is still challenging to develop drugs to precisely intervene in the IRF5 pathway. This review deeply discusses the important role of IRF5 in regulating immune cell function, and systematically reviews the existing intervention methods targeting IRF5, in order to provide some reference for the development of IRF5 inhibitors for the treatment of immune diseases in the future.
