参与P-gp膜定位和功能调控的相关蛋白研究进展

Research Progress of Proteins Involved in the Location and Function Regulation of P-gp on Plasma Membrane

  • 摘要: P-糖蛋白(P-gp)作为ABC转运体家族的成员之一,只有定位在细胞膜上才具有外排药物及毒物的功能。P-gp在细胞膜上的定位和功能受到多种机制调控。P-gp通过ERM(ezrin/radixin/moesin)蛋白家族锚定在膜上,其中脂筏区域在P-gp转运通道的形成中起重要作用,微囊蛋白-1(Cav-1)定位于小窝,作为脂筏上一种重要蛋白成分,与P-gp结合调节其结构和功能。综述参与P-gp膜定位和功能调控的相关蛋白研究进展,以期为P-gp在膜上的功能和表达研究提供新的方向。

     

    Abstract: P-glycoprotein (P-gp), as a member of the ABC transporter family, possesses the function of pumping drugs and toxicity out of cells when located on the plasma membrane. This process is regulated by various factors. P-gp is anchored to the cell plasma membrane through the combination effect of the ERM (ezrin/radixin/moesin) protein family. The lipid raft region plays an important role in the formulation of the P-gp transport function on plasma membrane. Caveolin-1 (Cav-1) is located on the caveolae as an important function protein on lipid raft, which combines with P-gp to regulate its structure and function. The article reviews the research progress of proteins involved in the location and function regulation of P-gp on plasma membrane, so as to provide a new direction for the research on the function and expression of P-gp on plasma membrane.
