选择性/靶向性一氧化氮释放策略的研究进展

Advance in Selective/Targeted Nitric Oxide Releasing Strategies

  • 摘要: 一氧化氮(NO)作为气体信使和(或)效应分子,在心血管、神经、免疫等系统中发挥重要的生理和病理功能,其相关药物研究已成为医药研究的一个重要方面。鉴于NO广泛的生物活性,利用其产生某种治疗作用的难点在于如何选择性/靶向性地将NO释放至病变部位,发挥其治疗作用的同时抑制或减少其不良反应。针对这一关键科学问题,从局部给药策略、特定条件激活的NO供体分子、NO-纳米递送系统以及抗体-NO偶联等方面,综述选择性/靶向性NO释放策略的研究进展。

     

    Abstract: Nitric oxide (NO), as a gas messenger or effector molecule, plays an important physiological and pathological role in the cardiovascular, nervous, and immune systems. Research on NO-related drugs has become an important aspect of medical research. In view of the extensive biological activity of NO, the key issue in using NO to produce a certain therapeutic effect lies in how to selectively and targetedly release NO to the diseased site, generating therapeutic effects and reducing side effects. In response to this key scientific issue, this article reviews the advance in selective/targeted nitric oxide releasing strategies including topical administration, specific condition-activated NO donor molecules, NO-nano delivery systems, and antibody NO conjugation.
