基于蛋白质结构和动力学的别构分子理性设计研究——北京大学分子设计实验室近年研究进展

Protein Structure and Dynamics-based Rational Design of Allosteric Modulators——Recent Progress of Molecular Design Laboratory of Peking University

  • 摘要: 别构药物具有高选择性和抗耐药性的优势。相对于正构药物较为成熟的研究策略,目前别构调控分子的发现存在很大挑战。总结笔者实验室在别构调控研究领域取得的进展,包括别构位点的理论计算预测方法、别构调控分子理性设计的成功示例,并讨论了别构药物研发领域面临的挑战以及新的发展动向。

     

    Abstract: Allosteric drugs have the advantages of better selectivity and anti-drug-resistance. Compared with the orthosteric drugs with mature research strategies, discovering allosteric modulators remains more challenging. In this review, the progress of allosteric regulation study in the authors' laboratory is summarized, including computational prediction methods of allosteric sites, and several successful examples of rational allosteric modulator discovery. Major challenges and new trends in allosteric drugs discovery are also discussed.
