Research Advances on Tumor Microenvironment Responsive Nano-Preparations

  • 摘要: 肿瘤微环境响应型纳米制剂由于能够在靶器官选择性释药,促进肿瘤细胞摄取,已成为癌症治疗的有效手段。肿瘤微环境中的酸性环境、酶、细胞质和细胞核中的谷胱甘肽等均可被用作为内源性的刺激,使得药物在肿瘤组织或细胞中响应性释放,改善药物的组织分布、降低药物的副作用。围绕对肿瘤微环境具有pH响应、酶响应、温度响应和还原响应的纳米制剂,综述其基本原理、设计方法及研究进展,为抗肿瘤纳米制剂的开发提供参考。

     

    Abstract: Tumor microenvironment responsive nano-preparations enhancing tumor cellular uptake and triggering drug release at the target site have emerged as one of the most effective strategies for cancer treatments. The tumor microenvironment includes slightly acidic environments, matrix metalloproteases, lysosomal enzymes, high glutathione levels in the cytoplasm and cell nucleus, which have been used as the potential internal stimuli to activate responsive release in the tumor sites or cancer cells. This method can improve the biodistribution and reduce the side-effects of active drugs. This review highlights the mechanism, design and recent developments of pH, enzyme, reduction and thermal responsive nano-preparations so as to provide reference information for further development of anticancer nano-preparations.
