Research Progress of Single-Cell Protein Detection Techniques Based on Immunoassay

  • 摘要: 单细胞分辨率下解析蛋白质表达,对探究细胞异质性、肿瘤微环境及推进临床精准医学等基础生物学研究具有重要意义。伴随材料化学、核酸化学、蛋白质化学的进步,以及新型检测设备与数据处理系统的迭代升级,单细胞蛋白质检测方法快速发展。其中,基于免疫分析法的单细胞蛋白质检测技术,凭借良好的靶向性和广泛的适用性,成为单细胞蛋白质分析的重要手段。综述了基于免疫分析法的单细胞蛋白质检测技术的原理、优缺点及应用,并对单细胞蛋白质分析技术的发展趋势进行了展望。

     

    Abstract: Analyzing protein expression at the single-cell resolution is of crucial significance for exploring cellular heterogeneity and tumor microenvironment, and advancing basic biological research such as clinical precision medicine. With the advancement of material chemistry, nucleic acid chemistry, protein chemistry, as well as the iterative upgrading of new detection equipment and data processing systems, single-cell protein detection techniques have developed rapidly. Among them, single-cell protein detection techniques based on immunoassay, with their good targeting ability and wide applicability, have become important means for single-cell protein analysis. This article reviews the principles, advantages and disadvantages, as well as the applications of single-cell protein detection techniques based on immunoassay, with a prospect of the development trends of single-cell protein analysis technology.
