天然产物靶标识别策略的研究进展

Research Progress of Target Identification Strategies for Natural Products

  • 摘要: 天然产物因具有广泛的生物活性,一直是新药研发的重要来源。新药研发的关键环节包括药物筛选及作用蛋白靶标的识别。药物分子与靶标的相互作用是发挥药理活性的基础,明确天然产物的作用靶标,对阐明其分子机制并推动临床预防与治疗应用具有重要意义。随着生命科学技术、生物信息学及人工智能的快速发展,药物靶标识别方法不断推陈出新并持续优化,主要分为两类:一类是直接评估药物与靶标相互作用的直接筛选法;另一类是基于药物诱导后表型和信号通路变化进行分析的间接筛选法。这些方法相辅相成,共同为天然产物靶标鉴定提供了有力工具。综述当前多种常用的天然产物靶标识别方法,以期为后续天然产物药物研究提供参考。

     

    Abstract: Natural products, due to their wide range of biological activities, have always been an important source for new drug development. The key steps in new drug research and development include drug screening and the identification of protein targets. The interaction between drug molecules and targets serves as the basis for exerting pharmacological activities, so clarifying the action targets of natural products is of great significance for elucidating their molecular mechanisms and promoting their application in clinical prevention and treatment. With the rapid development of life science and technologies, bioinformatics, and artificial intelligence, methods for identifying drug targets have been constantly updated and optimized, being divided mainly into two categories: the direct screening method for directly assessing the interaction between drugs and targets, and the indirect screening method for analyzing the changes in phenotypes and signaling pathways induced by drugs. These methods complement each other and together provide powerful tools for the identification of targets of natural products. This article reviews a variety of commonly used methods for identifying the targets of natural products, with the aim of providing some reference for subsequent research on drugs from natural products.
