缓释微针的设计及生物医学应用

The Design and Biomedical Applications of Microneedles with Sustained Release

  • 摘要: 微针是由微米级的针头阵列组成的新型经皮给药方式,其能够以微创的方式穿透角质层屏障,将药物高效地递送至真皮层。目前微针大多数采用速释聚合物材料作为基质,当微针刺入皮肤后,微针基质材料迅速溶解释放药物,难以起到缓释作用,不适于长效给药或治疗慢性疾病。缓释微针可以将药物保留在生物降解型基质中缓慢释放,从而达到长效治疗疾病的目的,并且能够降低给药频率从而提高患者依从性。综述首先归纳了主要的缓释微针设计类型,并总结了缓释微针在生物医学领域的应用,如长效避孕、长效脱发治疗、长效抗菌、长效缓解术后疼痛及抗肿瘤等,最后对缓释微针的未来研究前景进行了展望。

     

    Abstract: Microneedles composed of micron-sized needle arrays are a novel transdermal drug delivery strategy with minimally invasive penetration through the stratum corneum barrier, thus efficiently delivering drugs to the corium layer. Currently, most microneedles use immediate release polymer materials as the matrix, which rapidly dissolves to release the drug without achieving sustained release, making it unfit for long-term drug delivery or treatment of chronic diseases. Sustained-release microneedles can retain the drug in the biodegradable matrix for slow release, with prolonged therapeutic effects and reduced dosing frequency, thus helping to enhance patient compliance. This review categorizes major designs of sustained-release microneedles, and summarizes the biomedical applications of sustained-release microneedles, including long-term contraception, long-acting treatment for hair loss, prolonged antimicrobial effects, long-term relief of postoperative pain and treatment of tumor, with a prospect of future research directions in sustained-release microneedles technology.
