Advances in Research on the Mechanisms of GABAergic Signaling in Tumors and Its Targeted Intervention Strategies

  • 摘要: γ-氨基丁酸(γ-aminobutyric acid,GABA)是神经系统中重要的抑制性神经递质,不仅参与调控正常生理功能,还与多种疾病的发生发展相关,在结直肠癌、肺癌、乳腺癌、黑色素瘤等多种肿瘤中扮演重要角色。肿瘤自身或机体其他细胞的GABA代谢紊乱可导致GABA浓度改变,进而直接作用于肿瘤细胞,或通过影响肿瘤微环境及免疫细胞间接调控肿瘤进程。研究表明,通过激活GABA的A型受体(GABAAR)或B型受体(GABABR),GABA可影响钙离子通路、磷脂酰肌醇3-激酶(phosphoinositide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)通路、cAMP反应元件结合蛋白(c AMP response element-binding protein,CREB)通路、丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)/细胞外信号调节激酶(extracellular signal-regulated kinase,ERK)/CREB通路、核因子κB(nuclear factorκB,NF-κB)通路、Wnt/β-catenin通路等关键信号通路,最终发挥促癌或抑癌的作用。干预GABA能信号已成为癌症治疗领域一个有潜力的新方向。综述了GABA能信号在肿瘤中的作用机制及其靶向干预策略的研究进展,以期为抗肿瘤新药研发提供思路与参考。

     

    Abstract: γ-Aminobutyric acid(GABA) is an important inhibitory neurotransmitter in the nervous system. It not only participates in regulating normal physiological functions but is also associated with the pathogenesis of various diseases, and plays a significant role in multiple tumors, including colorectal cancer, lung cancer, breast cancer, and melanoma. Disorders of GABA metabolism in tumor cells or other cells of the body can lead to alterations in GABA concentration, thereby directly affecting tumor cells, or indirectly regulating tumor progression by affecting the tumor microenvironment and immune cell activity. Studies have shown that by activating GABA type A receptors(GABAAR) or type B receptors(GABABR), GABA influences key signaling pathways such as calcium pathway, phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT) pathway, cAMP response element-binding protein(CREB) pathway, mitogen-activated protein kinase(MAPK)/extracellular signal-regulated kinase(ERK)/CREB pathway, nuclear factor κB(NF-κB) pathway, and Wnt/β-catenin pathway, ultimately exerting pro-tumorigenic or anti-tumorigenic effects. Intervention in GABAergic signaling pathway has emerged as a promising new direction in the field of cancer therapy. This review summarizes the advances in research on the mechanisms of GABAergic signaling in tumors and its targeted intervention strategies, aiming to provide insights and references for the development of novel anti-tumor drugs.
