The Role of Neuroinflammation in Amyotrophic Lateral Sclerosis and the Research and Development of Related Drugs

  • 摘要: 肌萎缩侧索硬化(amyotrophic lateral sclerosis,ALS)是一种以大脑和脊髓运动神经元进行性丢失为特征的致死性神经退行性疾病,其发病机制尚未完全阐明。越来越多的证据表明,神经炎症在ALS的病理进程中发挥着重要作用。综述了小胶质细胞、星形胶质细胞和外周免疫细胞等参与神经炎症的关键细胞在ALS疾病进程中的作用,并总结了靶向ALS神经炎症的药物临床研发进展,以期为ALS的病理研究和相关药物研发提供新的方向。

     

    Abstract: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive loss of motor neurons in the brain and spinal cord. Currently, causative pathogenic mechanisms in ALS remain poorly understood. Accumulating evidence suggests that neuroinflammation plays an important role in the pathological mechanisms of ALS. This article reviews the role of key cells involved in neuroinflammation during the pathogenesis of ALS, including microglia, astrocyte and peripheral immune cells, and summarizes the clinical development of drugs targeting neuroinflammation of ALS, aiming to provide new directions for the pathological research on ALS and the research and development of related drugs.
