Research Progress on the Regulatory Mechanisms of Osteosarcoma Cancer Stem Cells and Related Therapeutic Strategies

  • 摘要: 骨肉瘤是常见的原发性恶性骨肿瘤之一,主要发生于儿童和青少年。尽管手术联合化疗等综合治疗在局部控制方面取得一定进展,但转移性和复发性骨肉瘤的预后仍不理想。近年来,肿瘤干细胞(cancer stem cell,CSC)被认为在骨肉瘤的发生、进展、耐药及转移过程中发挥关键作用,成为研究热点。系统综述了骨肉瘤中CSC的生物学特性、关键分子标志物、信号通路调控机制及其与肿瘤微环境的相互作用,并探讨其作为临床治疗潜在靶点的转化价值,旨在为骨肉瘤的精准治疗提供理论依据和研究方向。

     

    Abstract: Osteosarcoma is one of the common primary malignant bone tumors and mainly occurs in children and adolescents. Although comprehensive treatments such as surgery combined with chemotherapy have achieved certain progress in local control, the prognosis of metastatic and recurrent osteosarcoma remains unsatisfactory. In recent years, cancer stem cells(CSCs) have been considered to play a key role in the occurrence, progression, drug resistance, and metastasis of osteosarcoma, and have become a research hotspot. This article systematically reviews the biological characteristics, key molecular markers, signal pathway regulatory mechanisms of CSCs in osteosarcoma and their interaction with the tumor microenvironment, and explores their translational value as potential targets for clinical treatment, aiming to provide a theoretical basis and research direction for the precise treatment of osteosarcoma.
