Abstract: Liver cancer is one of the most prevalent cancers worldwide, and its high rates of recurrence and mortality pose significant challenges in clinical practice. Cancer stem cells(CSCs) plays a pivotal role in therapeutic resistance in liver cancer. In recent years, research on targeted therapies against liver cancer stem cells(LCSCs) has provided new avenues for overcoming these obstacles. This review introduces the cellular origins and characteristics of LCSCs. On this basis, it summarizes recent advances in therapeutic strategies targeting LCSCs: targeting key signaling pathways, targeting surface markers, and targeting the tumor microenvironment(TME). It also discusses the challenges and future perspectives in this field, aiming to provide a theoretical reference for precision therapies in liver cancer.
